چکیده

Mature T-cell and natural killer (NK)-cell neoplasms (Table 1) represent a heterogeneous spectrum of post-thymic T-celland NK-cell-derived malignancies. They represent 12% of all nonHodgkin lymphomas (NHLs). Significant geographic variation is noted with increased prevalence of select subtypes [i.e. adult T-cell leukemia/lymphoma (ATLL) and extranodal NK/T-cell lymphoma, nasal type] seen in Asian nations that may reflect exposure to etiologic agents such as human T-cell leukemia virus-1 (HTLV-1) and Epstein–Barr virus (EBV). The pathologic assessment of these cancers can be challenging and often requires immunophenotypic, cytogenetic and/or molecular analysis to ensure appropriate classification (Table 2). The most common entities will be discussed, highlighting clinical and pathologic manifestations, prognostic features and therapeutic strategies.

T-cell and NK-cell posttransplantation lymphoproliferative disorders.

Posttransplantation lymphoproliferative disorders (PTLDs) of T-cell or natural killer (NK)-cell origin are an uncommon heterogeneous group of lymphoid proliferations that fulfill the criteria for one of the T- or NK-cell lymphomas/leukemias. This report summarizes 130 T/NK-cell PTLDs reported in the literature or presented at the Society for Hematopathology/European Association for Haematopatho...

Are T-LGL Leukemia and NK-Chronic Lymphoproliferative Disorder Really Two Distinct Diseases?

Mature Large Granular lymphocytes (LGL) disorders include a spectrum of conditions, ranging from polyclonal to clonal indolent and/or overt leukemic LGL proliferations. Most cases are represented by clonal expansions of TCRα/β+ LGL displaying a CD8+ phenotype with expression of cytotoxic T-cell antigens (CD57, CD16, TIA-1, perforin and granzyme B). Proliferations of CD3-CD16+ NK cells with a re...

The immunophenotypic attributes of NK cells and NK-cell lineage lymphoproliferative disorders.

Natural killer (NK) cells are a distinct lymphocyte subset that, like cytotoxic T cells, recognize and destroy abnormal self-cells. Unlike T cells, however, NK cells lack a uniquely rearranged antigen receptor gene or a single defining immunophenotypic attribute. Furthermore, there is considerable functional and phenotypic overlap between NK cells and cytotoxic T cells, and the latter are induc...

Clinical Features, Pathogenesis, and Treatment of Large Granular Lymphocyte Leukemias

Large granular lymphocytes (LGLs) are large lymphocytes with azurophilic granules in their cytoplasm. LGLs are either natural killer (NK) cells or T lymphocytes. Expansions of the LGLs in the peripheral blood are seen in various conditions, including three clonal disorders: T-cell LGL (T-LGL) leukemia, chronic lymphoproliferative disorders of NK cells (CLPD-NK), and aggressive NK-cell leukemia ...

Indolent T- and NK-cell lymphoproliferative disorders of the gastrointestinal tract: a review and update.

Primary gastrointestinal (GI) T- and NK-cell lymphomas are usually aggressive neoplasms associated with high morbidity and mortality. Over the past two decades, however, cases of primary GI lymphoproliferative disorders (LPDs) or lymphomas of T- or NK-cell derivation with indolent behavior have been reported. These LPDs are rare and they can be challenging to diagnose as they share clinical and...
